Reservoir solution is spotted first, followed by a second pass delivering the protein solution; both set to deliver a nominal 40 nl volume. Owing to a more minimal buffer, the protein component is delivered more rapidly (less variability in focusing the transducer position) and is likely to deliver a slightly larger volume than the more concentrated well solutions at a given fixed energy.
